Introduction of fluorescence microscopy has made gigantic progress in biology and medicine in the past several decades. Confocal microscopy enables 3D imaging with optical sectioning, and two-photon excitation fluorescence microscopy, with the advent of pulsed laser, provides high contrast as well as natural depth discrimination capability. Despite their versatility in optical imaging field, in-vivo cellular imaging and real-time diagnosis by tissue imaging cannot be implemented due to the imaging speed restricted by opto-mechatronics.
In addition, in-vitro molecular level interactions in the cell cannot be optically observed because of optical resolution limited by diffraction. Therefore, overcoming slow imaging speed and low optical resolution by manipulating laser illumination and detection schemes is a key issue in the optical microscopy instrumentation.
In this presentation, basic principles of fluorescence microscopy will be explained and current limitations on laser optical microscopy will be briefly discussed. And the following scientific findings or technologies for various applications in the medical and life sciences at different research levels, such as molecular, cellular and tissue levels will be introduced, with a focus
on the studies done as well as on-going research in our group:
- Designing new opto-mechanical actuators with changing geometrical optics in the microscopy
- Manipulating photon detection schemes spatially and temporally in the photodetector
- Using specific light-matter interactions during the imaging
- Utilizing wave optics for the illumination schemes in the microscopy

6. 연사약력 :
Daekeun Kim received B.S. and M.S. in Mechanical Design and Production Engineering from
Seoul National University in 1993 and 1995, respectively. He served as Lecturer in Mechanical Engineering, Korea Air Force Academy from 1995 to 1998. As a Ph.D. student at Massachusetts Institute of Technology, he worked on a development project on regular basis monitoring system for the elderly who have congestive heart failure from 1999 to 2002.
He has been a member of Biointrumentation Engineering Analysis and Microscopy (BEAM)group since 2002 and is currently a Ph.D. candidate in Mechanical Engineering, Massachusetts Institute of Technology. His research interests are opto-mechatronics and optical instrumentation applications in biology and medicine.
