Real-world biological samples require extensive pretreatmentsuch as desalting and preconcentration before they can be analyzed in a miniaturized device.This presentation will address a novel method to integrate protein(micro)dialysis and preconcentration on microchips. We developed a unique technique to laser-pattern nanoporous polymer membranes in microchip using a projection lithography and phase separation polymerization. We can engineer the membrane size, shape and the pore size.


For microdialysis, we built two mass exchangers in fused silica microchip.Both have 35 mm thick membranes fabricated inside 5 mm long microchannel, but one has membranes with molecular weight cutoff (MWCO of 2kD, and the other with 100kD. Results indicate that the mass exchanger with 2kD MWCO membranes can be used for desalting protein solutions while the mass exchanger with 100 kD can be used to separate cytosolic proteins from cell membrane fragments based on size.


For preconcentration we cast 50 mm thick membrane at the junction of a simple cross channel. Proteins electrophoretically stack at the membrane surface upon applying electric field across the membrane. Results show that the protein concentration increases at least by two orders of magnitude and that the increase is linearly proportion to concentration time and electric field strength applied. This is the first sample preconcentrator using porous media that shows the linearity complying to the theory.
